摘要:用马齿苋的幼嫩叶片和茎段建立马齿苋组织培养及植株再生体系。最适愈伤组织诱导的培养基为MS + 6-BA 2mg/L + 0.7%琼脂 + 3%蔗糖,培养时PH值为5.8。将生长良好的愈伤组织块转入分化培养基中诱导不定芽的分化,最适分化培养基为MS + 2mg/L 6-BA + 1.0mg/L NAA。再生苗在MS + NAA 0.1mg/L的培养基上诱导生根,在适宜温度25℃的培养室里经过15d的培养,100%生根成苗。利用不同浓度的NaCl(0,100,150,200,250mol/L)对马齿苋的种子和幼嫩叶片进行处理,筛选耐盐存活植株。结果表明在NaCl胁迫条件下,马齿苋表现一定的耐盐性,马齿苋种子的萌发和叶片诱导形成愈伤组织会受到影响,随着浓度的增加,抑制越明显。

Abstract: The tissue culture system of Portulaca oleracea L. with tender leaves and shoots was established. The optimal callus induced medium was MS + 6-BA 2 mg/L. The optimal shoot induced medium was MS + 6-BA 2.0 mg/L + NAA 1.0 mg/L. The differentiated shoots were cultured on the medium of MS + NAA 0.1 mg/L for 15 days and all the shoots could grow into plantlet. Using different concentrations of NaCl (0,100,150,200,250 mol / L) on seeds and tender leaves of purslane to salt stress, salt-tolerant plant were screened in this paper. The results show that when stressed by NaCl, Portulaca oleracea L. adapted to saltmorphic circumstances ,but the germination of purslane seeds and the formation of callus was inhibited. With increasing NaCl concentration, the degree of inhibition increased.
