摘要:进行了土样的化合物预处理试验,结果表明,抑制剂对土壤中微生物的选择性分离比较适宜的方法为:用土壤悬浮液(10-1)于40℃和pH为7的条件下,加6%酵母膏+0.05%SDS(5mmol/L磷酸缓冲液)震荡20min,, 水稀释至10-2,然后用平板稀释法接菌在高氏合成1号培养基和甘油天门冬酰胺琼脂的平板培养基上。1号土壤高氏培养基上酵母膏+SDS处理放线菌比空白处理提高了0.5%,2号土壤高氏培养基上酵母膏+SDS处理放线菌比空白处理提高了2.6%,1号土壤甘油天门冬酰胺培养基上酵母膏+SDS处理放线菌比空白处理提高了4.4%,2号土壤甘油天门冬酰胺培养基上酵母膏+SDS处理放线菌比空白处理提高了14.3%,表明酵母膏和0.05%的SDS(十二烷基硫酸钠)有刺激放线菌生长的作用.

Abstract :  The soil sample pretreatment test .The result show that inhibitors on the soil of microbes in selective separation is suitable method as follows : firstly ,collecting soil samples in the 120. C, secondly, deal with 1 h and 6% yeast cream and 0.05% SDS (5mmol/Lphosphate buffer) ,thirdly, shake 20 minutes .Lastly, I use water dilute to10-3 in 50. C and  on the condition that pH=7 ,and 6% specially designed 0.05% SDS (5mmol/L phosphoric acid buffer) concussion 10 minutes, I use water dilute to 10-2,then use plate dilution  method by  bacteria  in  high's synthesis 1 AGAR , usable and glycerin was flat the culture  medium. 1 the soil's medium high yeast anointed + SDS treatment than blank processing were improved by 0.5. 2 the soil's medium high yeast anointed + SDS treatment than blank processing were increased by 2.6%, the no. 1 soil glycerin culture medium yeast was usable anointed + SDS treatment than blank processing were improved by 4.4%.2 soil glycerin culture medium yeast was usable anointed + SDS treatment than blank processing were up 14.3%, showed that 0.05% of the cream and yeast SDS (sodium dodecyl sulfate) have exciting actinomyces the role of growth.
