摘  要

 

   本试验以白花蛇舌草作为供试材料,采用KT、2,4-D、蔗糖,三物质不同浓度和光质配比处理,通过四因素三水平正交试验设计,以筛选出适合白花蛇舌草愈伤组织继代培养的最佳配方,然后以最佳配方为基础,采用L9(34)试验设计He-Ne激光不同辐射剂量、不同辐射时间及不同光照强度的配比处理,通过对吲哚乙酸氧化酶活性等生理生化指标测定,筛选出有利于白花蛇舌草愈伤组织生长的最优组合。试验结果表明:在正常光培养条件下,白花蛇舌草的继代培养的最佳配方是MS+2,4-D4.0mg/L+KT1.5mg/L+蔗糖30g/L。在He-Ne激光辐射时间与剂量分别为3mA、3min和光照强度为1500lx时,对愈伤组织的吲哚乙酸氧化酶活性的影响达到最低;在He-Ne激光辐射时间与剂量分别为6mA、3min和光照强度为2000lx时,对愈伤组织的可溶性蛋白质含量的影响达到最大;在He-Ne激光辐射时间与剂量分别为9mA、3min和光照强度为2500lx时,对可溶性糖含量的影响达到最大。其中He-Ne激光辐射时间与剂量分别为3mA、3min和光照强度为1500lx时,为最佳组合。

ABSTRACT

 

   This experiment with Hedyotis diffusa willd as selected materials, Using KT、2,4-D、sucrose,Three different concentrations and light qualitative material ratio processing , Through four factors of three levels orthogonal experiment design, To screened for Hedyotis diffusa willd the best prescription of callus subculture, Then based on the best formula, Adopt L9 (34) experiment design He-Ne laser different radiation dose、different time and different illumination intensity radiation ratio of processing, Based on such indoles acetic acid enzymatic activity determination of physiological indexes, Screened out callus growth is conducive to the optimal combination of Hedyotis diffusa Willd. The results show that: Culture conditions in normal light, Hedyotis diffusa Willd subculture best formula is MS +2,4-D4 .0 mg / L + KT1.5mg / L + sucrose 30g / L. In the He-Ne laser irradiation time and dose of 3mA, 3min, and when the light intensity was 1500lx, Indole acetic acid callus of enzyme activities to meet the minimum;In the He-Ne laser irradiation time and dose of 6mA, 3min, and when the light intensity was 2000lx, For callus soluble protein content reached the maximum; In the He-Ne laser irradiation time and dose were 9mA, 3min, and when the light intensity was 2500lx, the maximum of Soluble sugar content. Which He-Ne laser irradiation time and dose of 3mA, 3min, and when the light intensity was 1500lx, For the best combination.
